Synapse Development Analysis

Synapse development analysis is the study of how neurons form, mature, and refine synaptic connections, providing insight into the organization and function of neural circuits. It examines processes such as axon targeting, synaptic contact formation, assembly of pre- and postsynaptic components, neurotransmitter signaling, and activity-dependent remodeling across development. Researchers combine microscopy, molecular labeling, electrophysiology, and quantitative image analysis to measure synapse number, structure, connectivity, and functional strength. These analyses help explain how precise circuits emerge, identify cellular mechanisms underlying neurodevelopmental disorders, and evaluate how genetic changes, environmental factors, or therapeutic interventions affect neural connectivity.

The Synapse

Neurons communicate with one another by passing on their electrical signals to other neurons. A synapse is the location where two neurons meet to exchange signals. At the synapse, the neuron that sends the signal is called the presynaptic cell, while the neuron that receives the message is called the postsynaptic cell. Note that most neurons can be both presynaptic and postsynaptic, as they both transmit and receive information. An electrical synapse is one type of synapse in which the pre-...

Quantifying Synapses: an Immunocytochemistry-based Assay to Quantify Synapse Number

This protocol details how to quantify synapse number both in dissociated neuronal culture and in brain sections using immunocytochemistry. Using compartment-specific antibodies, we label presynaptic terminals as well as sites of postsynaptic specialization. We define synapses as points of colocalization between the signals generated by these markers.

Qualitative and Quantitative Analysis of the Immune Synapse in the Human System Using Imaging Flow Cytometry

Here, we describe a complete workflow for the qualitative and quantitative analysis of immune synapses between primary human T cells and antigen-presenting cells. The method is based on imaging flow cytometry, which allows the acquisition and evaluation of several thousand cell images within a relatively short period of time.

Microscopic Analysis of Synapses in Mouse Hippocampal Slices Using Immunofluorescence

Source: McLeod, F., et al. Evaluation of Synapse Density in Hippocampal Rodent Brain Slices. J. Vis. Exp. (2017).This video demonstrates a method for visualizing synaptic markers in hippocampal brain slices using confocal immunofluorescence analysis. The slices are immunostained with primary antibodies targeting presynaptic and postsynaptic markers, followed by fluorophore-conjugated secondary antibodies. The labeled markers are then visualized using confocal microscopy to evaluate the synaptic...
