Dopamine Agonists

Dopamine agonists are medications that activate dopamine receptors to mimic or enhance dopamine signaling, making them important tools in pharmacology and the treatment of disorders involving dopaminergic pathways. Direct agonists bind dopamine receptors, particularly D1-like or D2-like receptors, and trigger intracellular signaling, while some agents indirectly increase receptor stimulation by promoting dopamine release or reducing its breakdown. Clinicians use these drugs to manage Parkinson’s disease, hyperprolactinemia, and, in selected cases, restless legs syndrome. Their effects and adverse reactions, including nausea, low blood pressure, hallucinations, and impulse-control problems, reflect the widespread roles of dopamine in movement, endocrine regulation, reward, and cognition.

Creating Dynamic Images of Short-lived Dopamine Fluctuations with lp-ntPET: Dopamine Movies of Cigarette Smoking

We present a novel PET imaging approach for capturing dopamine fluctuations induced by cigarette smoking. Subjects smoke in the PET scanner. Dynamic PET images are modeled voxel-by-voxel in time by lp-ntPET, which includes a time-varying dopamine term. The results are 'movies' of dopamine fluctuations in the striatum during smoking.

Quantifying Agonist Activity at G Protein-coupled Receptors

A method for estimating the affinity constant of an agonist for the active state (Kb) of a G protein-coupled receptor is described. The analysis provides absolute or relative measures of Kb depending on whether constitutive receptor activation is measurable. Our method applies to various responses downstream from receptor activation.

Isolation and Culture of Primary Embryonic Mouse Midbrain Dopamine Neurons

The video demonstrates the process of isolating and culturing dopamine neurons from the midbrain of mouse embryos. The isolated ventral midbrain floors are dissociated using enzymes. The resulting dopamine neurons are harvested and transferred into micro-islands inside multi-well plates. The cells are incubated with a dopamine neuron medium to maintain the cells at a high density inside the micro-islands.
