This study explores a method for delivering small interfering RNA (siRNA) targeting prion protein expression in a mouse model. The approach utilizes RVG-9r peptide to facilitate transport across the blood-brain barrier, aiming to reduce neurodegenerative disease-related protein levels.
Targeted siRNA delivery to neurons addresses a critical bottleneck in neurodegenerative disease therapeutics by enabling precise gene silencing in the CNS. This approach supports mechanistic de-risking of prion protein as a therapeutic target through direct evidence of target engagement and pathway modulation. Successful demonstration of blood-brain barrier penetration via receptor-mediated transport enhances confidence in translating siRNA-based strategies to preclinical development pipelines.
The method fits within the early discovery to lead identification continuum by providing a tool for target validation and mechanistic insight generation prior to compound screening campaigns.
