Spectrometric Measurements

Spectrometric measurements are analytical techniques that quantify how matter interacts with electromagnetic radiation, revealing chemical composition, concentration, and physical properties. In practice, an instrument directs selected wavelengths or a broad spectrum of light through or onto a sample, then records transmitted, absorbed, emitted, or scattered radiation as a spectrum; characteristic signals can be compared with standards to identify and quantify analytes. In bioengineering, these measurements support protein and nucleic acid analysis, monitoring of enzymatic reactions, characterization of biomaterials, and evaluation of cell or tissue behavior. Their speed, sensitivity, and ability to use small samples make them valuable for research, diagnostics, and process control.

Here, we present detailed protocols for solid-state amide hydrogen/deuterium exchange mass spectrometry (ssHDX-MS) and solid-state photolytic labeling mass spectrometry (ssPL-MS) for proteins in solid powders. The methods provide high-resolution information on protein conformation and interactions in the amorphous solid-state, which may be useful in formulation design.

In-depth analyses of cancer cell proteomes facilitate identification of novel drug targets and diagnostic biomarkers. We describe an experimental workflow for quantitative analysis of (phospho-)proteomes in cancer cell subpopulations derived from liquid and solid tumors. This is achieved by combining cellular enrichment strategies with quantitative Super-SILAC-based mass spectrometry.

Mass Spectrometric Analysis of Glycosphingolipid Antigens

A specific and sensitive method to gain insight into the expression profile of glycosphingolipid antigens in immune organs and cells is described. The method takes advantage of the ion trap mass spectrometry allowing step-wise fragmentation of glycosphingolipid molecules for structural analysis in comparison to chemically synthesized standards.
