Cdna Microarray Processing

cDNA microarray processing is the laboratory and computational workflow used to measure the relative expression of many genes simultaneously. It converts messenger RNA into complementary DNA, labels samples with fluorescent dyes, and allows the labeled cDNA to hybridize to complementary probes fixed on a microarray. After scanning, signal intensities are background-corrected, normalized, and compared between experimental conditions to identify genes with altered expression. In biology, this process supports studies of development, disease mechanisms, drug responses, and cellular pathways, while careful quality control helps distinguish biological variation from technical noise.

The cDNA microarray PtGen2 was developed for gene expression studies in loblolly pine, P. taeda, and other conifer species. Here, we show pre- and post-hybridization handling and washing techniques that can be used with this array to yield better consistency, reduced artifacts, and lower backgrounds.
