Cell Culture Technique

Cell culture technique is the controlled growth and maintenance of cells outside an organism, providing a reproducible system for studying cellular structure, function, and behavior. In this method, cells are placed in a nutrient-rich culture medium and maintained under suitable conditions of temperature, pH, gas exchange, and sterility; adherent cells are periodically detached and passaged as they reach confluence. Cell culture supports research in molecular and developmental biology, disease modeling, drug testing, and biotechnology by enabling direct observation and experimental manipulation of living cells. Careful aseptic handling and consistent culture conditions improve reproducibility and help preserve cell viability and relevant biological characteristics.

Proximal Culture System: An In Vitro Culture Technique to Study Paracrine Signaling Between Cells

2023

In this video, we develop a proximal co-culture model of ovarian cancer cells and primary human mesothelial cells. This physiologically relevant model allows the study of paracrine signaling, where the localized concentrations of the secreted factors are maintained while preventing direct cellular contact.

A Co-culture Technique for Differentiating Human Neural Progenitor Cells into Neurons

2025

This video demonstrates a method to differentiate human neural progenitor cells (NPCs) into neurons. The NPCs are introduced onto an established mouse astrocyte-rat cortical neuron co-culture, promoting NPC differentiation into neurons. The differentiation is confirmed by visualizing the cells under a confocal microscope.

Eliminating specific cells without damaging other cells is extremely difficult, especially in established tissue, yet there is an urgent need for a cell elimination method in the tissue engineering field. Here, we present a method for specific cell elimination from a mixed 3D cell culture using near infrared photoimmunotherapy (NIR-PIT).

A Technique to Culture Mouse Hippocampal Neurons

2025

The video demonstrates a method for culturing neurons from murine hippocampi. Initially, the hippocampi undergo enzymatic digestion to break down the extracellular tissue matrix. Subsequently, the digested tissue is mechanically disrupted to isolate individual neurons. Finally, these neurons are cultured on glial feeder cells.
