Collagen Based Matrix

A collagen-based matrix is a three-dimensional biomaterial formed from collagen, the structural protein that supports tissues and influences cell behavior. Collagen molecules assemble into fibrils or hydrated gels that provide mechanical support, bind cell-surface receptors, and create an extracellular-matrix-like environment for cell adhesion, migration, and growth. In biology, these matrices are used to culture cells, model tissue architecture, study cell–matrix interactions, and develop scaffolds for tissue engineering and regenerative research. Their composition, concentration, crosslinking, and stiffness can be adjusted to reproduce specific tissue conditions, making them valuable for investigating development, disease processes, and repair.

Cell migration is an important part of human development and life. In order to understand the mechanisms that can alter cell migration, we present a planar gradient diffusion system to investigate chemotaxis in a 3D collagen matrix, which allows one to overcome modern diffusion chamber limitations of existing assays.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.

Liver diseases are induced by many causes that promote fibrosis or cirrhosis. Transplantation is the only option for recovering health. However, given the scarcity of transplantable organs, alternatives must be explored. Our research proposes the implantation of collagen scaffolds in liver tissue from an animal model.
