Definitive Endoderm

Definitive endoderm is one of the three primary embryonic germ layers, giving rise to the epithelial linings of the digestive and respiratory tracts and to organs such as the liver and pancreas. During gastrulation, strong Nodal or Activin signaling promotes mesendoderm formation and activates transcription factors including SOX17 and FOXA2, establishing definitive endoderm while suppressing alternative cell fates. In developmental biology, researchers use these molecular cues to guide pluripotent stem cells through staged differentiation into endodermal progenitors and specialized organ cells. This process supports studies of embryonic development, disease modeling, drug testing, organoid formation, and potential regenerative therapies.
