Dendritic Cell Culture

Dendritic cell culture is a laboratory method for growing and maintaining dendritic cells or their precursors under controlled conditions to study antigen presentation and immune regulation. In vitro, progenitor cells or monocytes are exposed to defined cytokines that support differentiation, while maturation stimuli can induce changes in surface molecules, cytokine secretion, and the ability to activate T cells. Researchers use these cultures to investigate innate and adaptive immunity, host responses to pathogens, tumor immunology, and vaccine development. The approach also provides a controllable model for evaluating how dendritic cells capture, process, and present antigens.

Culture of myeloid dendritic cells from bone marrow precursors

This video demonstrates the procedure for differentiating myeloid dendritic cells from mouse bone marrow. Isolation of mouse tibia and femur, and processing of bone marrow are demonstrated. Pictures demonstrating cell morphology before and after differentiation, and figures depicting cell phenotype and IL-12 production following maturation using CpG are shown.

We present a protocol that permits to view and to quantitatively asses the morphology of the dendritic tree of individual Purkinje cells grown in organotypic cerebellar slice cultures. This protocol is intended to promote studies on the mechanisms of Purkinje cell dendritic development.

T Cells Capture Bacteria by Transinfection from Dendritic Cells

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.

Numerous recent studies have identified mutations in synaptic proteins associated with brain pathologies. Primary cultured cortical neurons offer great flexibility in examining the effects of these disease-associated proteins on dendritic spine morphology and motility.
