Dermal Incision

Dermal incision is a deliberate cut through the skin, used to create access to underlying tissues or to study how biological systems respond to injury. A sharp instrument mechanically separates the epidermal and dermal layers, disrupting small blood vessels and extracellular matrix; the resulting wound initiates hemostasis, inflammation, cell migration, re-epithelialization, and collagen-based remodeling. In biology and biomedical research, controlled dermal incisions support investigations of wound healing, tissue regeneration, immune responses, and surgical repair, while standardized depth, length, and closure conditions help researchers compare healing outcomes across experiments.

Generating Neurons by Reprogramming Human Dermal Fibroblast Cells

2025

The video demonstrates the generation of neurons from adult human dermal fibroblasts (aHDFs). The aHDFs are reprogrammed using a lentiviral vector carrying genes for neuronal transcription factors and short hairpin RNAs (shRNAs). The shRNAs inhibit the expression of a neuronal transcriptional repressor, and the neuronal transcription factors activate genes to differentiate aHDFs into neurons. These reprogrammed aHDFs are cultured in an early and a late neuronal conversion medium to generate...

Murine Dermal Fibroblast Isolation by FACS

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Metacarpal Small Incision for Carpal Tunnel Syndrome

2024

Carpal tunnel decompression with transverse carpal ligament transection is a very successful treatment of carpal tunnel syndrome. The standard long-curved incision technique has become the best treatment for median nerve entrapment decompression. Here we introduce the metacarpal small incision carpal tunnel release (MSICTR) in the treatment of carpal tunnel syndrome (CTS).

This paper describes a method for magnetic bead-based isolation of murine endothelial cells from dermal lymphatic capillaries. The isolated lymphatic endothelial cells can be used for downstream in vitro experiments and protein expression analysis.

Whole Mount Preparation of Mouse Intestine: Using a Simple Gut-incising Device to Prepare Whole Mount

2023

In this video, we demonstrate whole mount preparation of the mouse intestine using a simple gut-incising device. The resulting mounts are of superior and uniform quality and can be used for histological analyses.
