Diamond Blade Sectioning

Diamond blade sectioning is a precision method for cutting hard biological specimens into thin, analyzable sections while preserving their structural organization. A rotating blade embedded with industrial diamond particles abrades materials such as bone, teeth, and mineralized tissue, while controlled cutting conditions and cooling help limit heat, vibration, and mechanical damage. The resulting sections can support histological staining, microscopy, and morphological analysis, including studies of tissue interfaces and implanted materials. In biology, this technique is especially valuable when conventional blades cannot penetrate rigid specimens without excessive deformation, enabling more accurate investigation of native architecture and pathology.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.
