Drosophila Neuronal Cultures

Drosophila neuronal cultures are laboratory preparations of fruit-fly neurons maintained outside the organism, providing a controlled system for studying nervous-system development and function. Researchers isolate and dissociate neural tissue, plate the cells in defined culture conditions, and monitor how they extend axons and dendrites, form synaptic connections, and respond to genetic or pharmacological manipulation. These cultures support analysis of neuronal differentiation, membrane excitability, synaptic signaling, and cellular responses to disease-associated genes or compounds. Combined with Drosophila genetics and imaging, they help connect molecular pathways to neuronal phenotypes and provide an efficient platform for investigating conserved mechanisms of neural biology.

This video demonstrates the preparation of primary neuronal cultures from midgastrula stage Drosophila embryos. Views of live cultures show cells 1 hour after plating and differentiated neurons after 2 days of growth in a bicarbonate-based defined medium. The neurons are electrically excitable and form synaptic connections.

Primary Neuronal Cultures

2023

The complexity of the brain often requires neuroscientists to use a simpler system for experimental manipulations and observations. One powerful approach is to generate a primary culture by dissecting nervous system tissue, dissociating it into single cells, and growing those cells in vitro. Primary cultures make neurons and glia easily accessible to the experimental tools required for techniques like genetic manipulation and time-lapse imaging. Furthermore, these cultures represent a highly...
