Early B Cell Igm Expression

Early B cell IgM expression is the developmental stage at which newly formed B lymphocytes first display membrane-bound immunoglobulin M (IgM) as their B-cell receptor (BCR), signaling successful assembly of the antigen-recognition machinery. During B-cell development, V(D)J recombination generates a functional immunoglobulin heavy-chain gene; the μ heavy chain pairs first with surrogate light chains in a pre-BCR and then with conventional light chains to produce surface IgM. This transition supports selection, survival, and maturation of B cells and establishes early immune surveillance. Measuring early B cell IgM expression helps investigate lymphocyte development, receptor signaling, immune deficiencies, and abnormal B-cell states in experimental and clinical biology.

Generation of Two-color Antigen Microarrays for the Simultaneous Detection of IgG and IgM Autoantibodies

We describe here a method to generate customizable antigen microarrays that can be used for the simultaneous detection of serum IgG and IgM autoantibodies from humans and mice. These arrays allow for high-throughput and quantitative detection of antibodies against any antigens or epitopes of interest.

We present a protocol to identify protein moieties containing antigens for human and mouse IgM antibodies with specific kappa (Vκ) light chains. This protocol is not limited to IgM class antibodies but applies to all immunoglobulin isotypes that target their antigen with sufficiently high affinity during immunoprecipitations.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.

Here we describe a method for retroviral overexpression and adoptive transfer of murine B-1a cells to examine in vivo B-1a cell migration and localization. This protocol can be extended for diverse downstream functional assays including quantification of donor B-1a cell localization or analysis of donor cell-derived secreted factors post-adoptive transfer.
