Embryonic Valve Cells

Embryonic valve cells are developing heart cells that form the tissue architecture of cardiac valves, which regulate one-way blood flow through the heart. During embryogenesis, selected endocardial cells undergo epithelial-to-mesenchymal transition, migrate into the endocardial cushions, and produce extracellular matrix that is remodeled into flexible valve leaflets. Their coordinated responses to mechanical forces and developmental signals shape valve structure and function. Studying these cells helps explain congenital valve malformations, valve maturation, and disease mechanisms, while supporting research on tissue engineering, regenerative therapies, and experimental models of cardiovascular development.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.

The ability of human embryonic stem cells to self-renew and differentiate into all cell types of the body suggests that they hold great promise for both medical applications and as a research tool for addressing fundamental questions in development and disease. Here, we provide a concise, step-by-step protocol for the derivation of human embryonic stem cells from embryos by immunosurgical isolation of the inner cell mass.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.
