Endocardial Cell Isolation

Endocardial cell isolation is the process of separating endocardial cells from cardiac tissue for controlled study of the heart’s inner lining and its signaling functions. The procedure typically combines tissue dissection with enzymatic digestion and gentle mechanical dissociation to release cells, followed by filtration, centrifugation, and culture under conditions that support cell viability and identity. Isolated endocardial cells enable investigations of cardiac development, valve formation, endothelial signaling, and responses to disease-related or pharmacological stimuli. By providing defined cellular models, this technique supports studies of heart biology and the development of experimental approaches for cardiovascular research.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.
