Endothelial Cell Branching

Endothelial cell branching is the process by which endothelial cells extend and organize into interconnected tubular networks, a key event in blood vessel formation and tissue vascularization. During angiogenesis, signals such as vascular endothelial growth factor stimulate endothelial cells to migrate, proliferate, and coordinate through tip-cell guidance, stalk-cell elongation, and lumen formation. Branching is shaped by cell-cell adhesion, extracellular matrix interactions, and local chemical gradients that determine vessel direction and network structure. Studying this process helps explain embryonic development, wound healing, tumor vascularization, and vascular disease, while branching assays provide models for evaluating angiogenic mechanisms and potential therapies.

Glycogen Branching Assay to Measure the Degree of Glycogen Branching

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.

Isolation of Embryonic Ventricular Endothelial Cells

Primary cell culture is a useful technique for analyzing specific populations of cells, particularly from transgenic mouse embryos at specific developmental stages. Herein, embryonic ventricles are dissected and dissociated, and antibody-conjugated beads recognize and separate out the endothelial cells for further analysis.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

Novel isolation methods of primary endothelial cells from blood vessels are needed. This protocol describes a new technique that completely inverts blood vessels of interest, exposing only the endothelial side to enzymatic digestion. The resulting pure endothelial cell culture can be used to study cardiovascular diseases, disease modelling, and angiogenesis.
