Epididymal Sperm

Epididymal sperm are spermatozoa that have left the testes and are undergoing, or have completed, functional maturation within the epididymis, a duct essential for male fertility. As sperm travel through its distinct regions, epididymal epithelial secretions and changing luminal conditions remodel the sperm membrane and surface proteins, enabling progressive motility and preparing them for fertilization after subsequent capacitation. The distal epididymis also stores mature sperm before ejaculation. Studying epididymal sperm helps explain post-testicular maturation, diagnose male infertility, evaluate sperm quality, and develop improved approaches to contraception and assisted reproduction.

Proteomic analysis of any cell type is highly dependent on both purity and pre-fractionation of the starting material in order to de-complexify the sample prior to liquid chromatography mass spectrometry (MS). By using back-flushing techniques, pure spermatozoa can be obtained from rodents. Following digestion, phosphopeptides can be enriched using TiO2.
