Erythroid Progenitor Identification

Erythroid progenitor identification is the process of recognizing and distinguishing immature blood-forming cells committed to red blood cell production. It is important for mapping hematopoietic development and evaluating disorders of erythropoiesis. In practice, researchers combine cell-surface marker profiles with flow cytometry, often using fluorescently labeled antibodies against markers whose expression changes as cells progress from early progenitors toward erythroblasts and mature erythrocytes. This approach can be complemented by cell morphology, colony-forming assays, and gene-expression analysis to validate lineage and developmental stage. Accurate identification supports studies of red blood cell formation, anemia, leukemia, stem cell differentiation, and regenerative or drug-testing models.

Detection of Residual Donor Erythroid Progenitor Cells after Hematopoietic Stem Cell Transplantation for Patients with Hemoglobinopathies

Quantification of donor-derived cells is required to monitor engraftment after stem cell transplantation in patients with hemoglobinopathies. A combination of flow cytometry-based cell sorting, colony formation assay, and subsequent analysis of short tandem repeats may be used to assess the proliferation and differentiation of progenitors in the erythroid compartment.

Here, we describe a novel flow cytometric method for prospective isolation of early burst-forming unit erythroid (BFU-e) and colony-forming unit erythroid (CFU-e) progenitors directly from fresh mouse bone marrow and spleen. This protocol, developed based on single-cell transcriptomic data, is the first to isolate all the tissue's erythroid progenitors with high purity.
