Fitc-dextran

FITC-dextran is a fluorescently labeled polysaccharide used as a tracer to visualize transport, permeability, and uptake in biological systems. It consists of dextran, a water-soluble glucose polymer, covalently linked to fluorescein isothiocyanate, whose fluorophore emits green light when excited at an appropriate wavelength. Because dextran size influences movement through barriers, researchers use FITC-dextran to measure membrane and epithelial permeability, assess vascular leakage, and track endocytosis or intracellular trafficking by fluorescence microscopy or plate-based assays. Molecular-weight variants support experiments ranging from intestinal barrier studies to blood-brain barrier models, helping quantify structural integrity and changes caused by disease, drugs, or experimental treatments.

This video demonstrates a method to study permeation through an in vitro blood-brain barrier (BBB) model using astrocytes and endothelial cells on a membrane insert, with free FITC and FITC-loaded ferritin. FITC-ferritin bound to receptors on endothelial cells and crossed the membrane barrier to the lower chamber while free FITC showed restricted permeation and remained in the upper chamber.
