Formulation Stabilization

Formulation stabilization is the process of maintaining a biological product’s physical, chemical, and functional properties throughout storage, handling, and use. It works by controlling conditions such as pH, temperature, moisture, and oxygen while selecting compatible excipients that reduce degradation, aggregation, precipitation, or loss of biological activity. In biology, stabilization supports the development of proteins, enzymes, vaccines, nucleic acids, and cell-based materials that remain consistent and effective over time. Effective formulations improve shelf life, product quality, and experimental reproducibility, while helping researchers and manufacturers preserve sensitive biological components during transport and application.

Freeze-drying is often an easy and convenient way to obtain dry products of viable bacterial cells. An issue of the process is cell survival. We detail here a procedure to investigate how cell survival during freeze-drying is influenced by the properties of the formulation used.

Evaluating the Effect of a Synbiotic Formulation on Alzheimer’s Disease Pathology in Drosophila

Take a transgenic Drosophila. The brain of the Drosophila expresses the human amyloid peptide protein or APP and a cleaving enzyme. The enzyme cleaves APP, enabling the formation of beta-amyloid plaque, which leads toAlzheimer's disease or AD pathology in Drosophila.Feed the Drosophila AD model a diet containing a synbiotic formulation. The formulation contains a combination of three bioactive probiotic bacteria and a prebiotic.Inside the fly's gut, the prebiotic supports the growth and survival...

Nuclear Stability

2020

Protons and neutrons, collectively called nucleons, are packed together tightly in a nucleus. With a radius of about 10−15 meters, a nucleus is quite small compared to the radius of the entire atom, which is about 10−10 meters. Nuclei are extremely dense compared to bulk matter, averaging 1.8 × 1014 grams per cubic centimeter. If the earth’s density were equal to the average nuclear density, the earth’s radius would be only about 200 meters. To hold positively charged protons together in the...

RNA Stability

Intact DNA strands can be found in fossils, while scientists sometimes struggle to keep RNA intact under laboratory conditions. The structural variations between RNA and DNA underlie the differences in their stability and longevity. Because DNA is double-stranded, it is inherently more stable. The single-stranded structure of RNA is less stable but also more flexible and can form weak internal bonds. Additionally, most RNAs in the cell are relatively short, while DNA can be up to 250 million...

Monitoring Bacterial Growth in Distinct Media Formulations with an Automated Microbioreactor

Source: Hemmerich, J., et al. Protocol for Optimization of Heterologous Protein Production Using Automated Microbioreactor Technology. J. Vis. Exp. (2017).This video demonstrates the use of an automated microbioreactor to monitor bacterial growth and fluorescent protein secretion in response to varying media formulations. A genetically modified bacterium expressing a secretory fluorescent protein is cultured under antibiotic selection and inoculated into nutrient-varied wells. Real-time optical...
