Genetic Knockout Analysis

Genetic knockout analysis is a research approach used to determine a gene’s function by disrupting or eliminating its activity and examining the resulting effects. A knockout can be created by targeted genome editing, such as introducing mutations that prevent a gene from producing a functional product, after which researchers compare modified cells or organisms with appropriate controls. Changes in growth, development, physiology, or behavior can reveal the gene’s biological role and interactions with other pathways. In biology, this analysis supports studies of gene function, disease mechanisms, developmental processes, and potential therapeutic targets.

An Overview of Genetic Analysis

An organism’s physical traits, or phenotype, are a product of its genotype, which is the combination of alleles (gene variants) inherited from its parents. To varying degrees, genes interact with each other and environmental factors to generate traits. The distribution of alleles and traits within a population is influenced by a number of factors, including natural selection, migration, and random genetic drift. In this video, JoVE introduces some of the foundational discoveries in genetics,...

Transgenesis, or the use of genetic engineering to alter gene expression, is widely used in the field of developmental biology. Scientists use a number of approaches to alter the function of genes to understand their roles in developmental processes. This includes replacement of a gene with a nonfunctional copy, or adding a visualizable tag to a gene that allows the resultant fusion protein to be tracked throughout development. In this video, the viewers will learn about the principles behind...

Recent development in gene targeting tools makes production of knockout (KO) rabbits possible. In the present work, we generated five Apolipoprotein (Apo) C-III KO rabbits using Zinc Finger Nucleases (ZFN). This work demonstrated that ZFN is a highly efficient method to produce KO rabbits.
