Germline Tissue

Germline tissue is the cell population that produces reproductive cells, or gametes, and transmits genetic information from one generation to the next. In animals, germ cells develop within specialized tissue environments, undergo mitosis to expand their numbers, and then enter meiosis, which reduces chromosome number and generates genetic variation before sperm or egg formation. Studying germline tissue helps explain fertility, inheritance, embryonic development, and the origins of reproductive disorders. Research in this area also supports investigations of reproductive aging, developmental biology, genetic disease, and how environmental conditions can influence heritable biological outcomes.

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.

This video describes microinjection, a common method of creating transgenic C. elegans strains. In the example, we will see microinjection used to introduce a ribonucleoprotein (RNP) complex for CRISPR-bases gene editing.

Mosquito Embryo Microinjection: A Technique to Deliver Exogenous DNA into Embryo Yolk of Anopheles Gambiae to Generate Germline Mutations

2025

In this video, we demonstrate the delivery of gene drive construct into the embryo of Anopheles gambiae mosquito through embryo microinjection technique. This procedure allows for integration of delivered DNA construct, through homologous recombination, into the germline cells to generate transformed mosquito population.
