Growth Chamber Incubation

Growth chamber incubation is a laboratory method for maintaining biological specimens under controlled environmental conditions, enabling researchers to study growth and development with greater precision and reproducibility. Chambers regulate variables such as temperature, light intensity, photoperiod, humidity, and sometimes carbon dioxide concentration to create defined conditions for organisms or cultures. In biology, this approach supports seed germination, plant physiology, microbial growth, and experiments on environmental responses. By isolating specific conditions and enabling consistent replication, growth chamber incubation helps distinguish biological effects from environmental variation and produces reliable data for research, agriculture, and biotechnology.

Microscopic Analysis of Bacterial Growth Under Stress Using Microfluidic Chambers

2025

Source: Cayron, J., et al. Multi-scale Analysis of Bacterial Growth Under Stress Treatments. J. Vis. Exp. (2019).This video demonstrates how to prepare and use a microfluidic plate for imaging bacterial cells under stress conditions. It outlines the process of introducing a bacterial culture and a stress-inducing antibiotic medium, followed by cell loading and microscopic setup. Time-lapse imaging in phase-contrast mode is then used to observe the development of elongated, filamentous cells...

Building and Operating a Low-Cost Elevated Carbon Dioxide Growth Chamber to Evaluate Microgreen Physiology under Spaceflight-Relevant CO2 Levels

2026

This protocol describes how to construct and operate a small, low-cost growth chamber capable of maintaining extreme carbon dioxide (exCO2) levels for plant experiments. The chamber enables reproducible studies of microgreen physiology under spaceflight-relevant atmospheric conditions, using a single-board computer-based monitoring and control system and a passive hydroponic wicking system.

A General Method for Evaluating Incubation of Sucrose Craving in Rats

Responding for food or drug-paired cues increases over the course of a period of abstinence and this may relate to an increased susceptibility to relapse behaviors. Here we detail a procedure for evaluating this "incubation of craving" in rats that have self-administered sucrose.

A Technique for the Prolonged Incubation of Neuronal Tissue of the Retina

2025

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
