Localization Precision

Localization precision is the degree to which a biological molecule’s position can be estimated reproducibly, making it a key measure for resolving structures beyond the diffraction limit. In fluorescence microscopy, detectors collect photons from a labeled molecule, and software fits the resulting point-spread function to estimate its center; more photons, lower background, and appropriate imaging conditions generally reduce positional uncertainty. High localization precision supports single-molecule tracking, super-resolution imaging, and analysis of molecular organization, enabling researchers to distinguish closely spaced features, quantify movement, and relate nanoscale distributions to cellular function.

In Situ Hybridization for the Precise Localization of Transcripts in Plants

The in situ hybridization protocol described here allows a direct localization of mRNA and small RNA expression at the cellular level with high sensitivity and specificity. The procedure is optimized for paraffin-embedded plant tissue sections, is applicable to a wide range of plants and tissues, and can be completed within ten days.

We demonstrate a method for grafting cultured cells into defined sites of early mouse embryos to determine their in vivo potential. We also introduce an optimized electroporation method that uses glass capillaries of known diameter, allowing the precise delivery of exogenous DNA into a few cells in the embryos.

Murine Appendectomy Model of Chronic Colitis Associated Colorectal Cancer by Precise Localization of Caecal Patch

The presented protocol describes a facile surgical removal of the appendix (caecal patch) in a mouse followed by the induction of inflammatory bowel disease-associated colorectal cancer. This murine appendectomy model enables investigation of the biological role of the appendix in the pathogenesis of human gastrointestinal disease.
