Mouse Infection Protocol

Mouse Infection Protocols are standardized procedures for introducing a pathogen or infectious agent into mice to study disease, host responses, and treatment effects in a controlled biological model. Researchers define the agent, inoculation route, dose, timing, and animal-monitoring criteria, then assess infection through clinical observations, pathogen measurements, tissue analysis, and immune readouts. These protocols help connect microbial virulence with host immunity and disease progression, supporting vaccine development, antimicrobial testing, and investigations of host-pathogen interactions. Carefully controlled procedures also improve reproducibility while meeting animal-welfare, containment, and experimental-design requirements.

Protocol for Dengue Infections in Mosquitoes (A. aegypti) and Infection Phenotype Determination

Once a gene is identified as potentially refractory for the dengue virus, it must be evaluated for it's role in preventing viral infections within the mosquito. This protocol illustrates how the extent of dengue infections of mosquitoes can be assayed. The techniques for growing up the virus in culture, membrane feeding mosquitoes human blood, and assaying viral titers in the mosquito midgut are demonstrated.

Once a gene is identified as potentially refractory for malaria, it must be evaluated for its role in preventing Plasmodium infections within the mosquito. This protocol illustrates how the extent of plasmodium infections of mosquitoes can be assayed. The techniques for preparing the gametocyte culture, membrane feeding mosquitoes human blood, and assaying viral titers in the mosquito midgut are demonstrated.

C. elegans has emerged as a new genetic model to study host-pathogen interactions. Here we describe a protocol to infect C. elegans with Salmonella typhimurium coupled with the double-strand RNAi interference technique to examine the role of host genes in defense against Salmonella infection.

Influenza A viruses (IAVs) are important human respiratory pathogens. To understand the pathogenicity of IAVs and to perform preclinical testing of novel vaccine approaches, animal models mimicking human physiology are required. Here, we describe techniques to evaluate IAV pathogenesis, humoral responses and vaccine efficacy using a mouse model of infection.
