Muscle Fatty Degeneration

Muscle fatty degeneration is the abnormal accumulation of fat within skeletal muscle, often accompanied by loss of contractile tissue, and is an important indicator of impaired muscle quality. It can develop after chronic injury, denervation, disuse, aging, or disease, when damaged muscle fibers are not fully regenerated and connective and adipose tissue expand between or within them. This remodeling reduces force production and may limit recovery, even when muscle size appears preserved. In biology and clinical research, magnetic resonance imaging, ultrasound, and histological analysis quantify fatty infiltration to assess conditions such as rotator cuff disease and muscular dystrophy, track progression, and evaluate rehabilitation or therapeutic strategies.

Immunolabelling Myofiber Degeneration in Muscle Biopsies

Described here is a protocol for direct immunolabelling of necrotic myofibers in muscle cryosections. Necrotic cells are permeable to serum proteins, including immunoglobulin G (IgG). Revealing the uptake of IgG by myofibers allows the identification and quantification of myofibers that undergo necrosis regardless of muscle condition.

We propose a protocol to identify fatty acids without the need to purify them. It combines information on the retention times with the mass spectra of three types of fatty acid derivatives: fatty acid methyl esters (FAMEs), 4,4-dimethyl oxazoline derivatives (DMOX), and 3-pyridylcarbinyl esters (picolinyl).
