Nuclei Gating

Nuclei gating is a flow cytometry strategy that identifies and selects isolated cell nuclei from a mixed sample, enabling focused analysis of nuclear features. The method uses light-scattering properties and fluorescence signals, often from DNA-binding dyes, to distinguish intact nuclei from debris, aggregates, and other particles; sequential gates can further isolate nuclei with the desired size, DNA content, or marker profile. In biology, nuclei gating supports cell-cycle analysis, ploidy measurement, genome-content assessment, and purification of nuclei for downstream molecular studies. Careful gate placement improves data quality by reducing contamination and separating distinct nuclear populations.
