Oviposition Assay

An oviposition assay is a biological method used to measure egg-laying behavior and reproductive preferences, commonly in insects and other egg-laying animals. In a typical assay, females receive one or more defined substrates or environmental conditions, and researchers quantify the number, location, or timing of eggs deposited; choice assays compare preferences, whereas no-choice assays assess acceptance of a single option. These measurements help evaluate how chemical cues, host characteristics, nutrition, temperature, or other factors influence reproduction. Oviposition assays support studies of animal behavior, ecology, host selection, pest management, and the development of strategies that alter or monitor reproductive outcomes.

This protocol describes a microinjection methodology that we have standardized and used for several years to deliver specific quantities of nucleic acids directly to the hemolymph of mosquitoes and house flies. This protocol results in minimal injection mortality and allows dose correlated measurements of fecundity.

Drosophila females choose between substrates and select sites that are attractive for egg-laying. This video describes a behavioral assay that tests their decision-making, called the egg-laying preference assay. The featured clip demonstrates the procedure with a custom-built setup, which enables researchers not only to test the behavior of many females at the same time but is also compatible with video recording and optogenetic setups.

Aortic Ring Assay

Angiogenesis, the sprouting of blood vessels from pre-existing vasculature, is associated with both natural and pathological processes. Here we demonstrate an aortic ring assay that allows angiogenic potentiators and inhibitors to be directly added to aortic rings in culture. Sprouting and neovessel outgrowth can be determined by inspecting the aortic rings over a period of 6-12 days.

Assaying DNA Damage in Hippocampal Neurons Using the Comet Assay

The comet assay is an efficient way of detecting single- and double-strand breaks, including alkali-labile sites and DNA-DNA/DNA-protein cross-links on the DNA in all cells including hippocampal neurons. The method takes advantage of the differential migration of DNA in an electric field due to differences in amount of DNA damage.
