Pancreatic Beta Cells

Pancreatic beta cells are specialized endocrine cells in the islets of Langerhans that regulate blood glucose by producing and releasing insulin. When glucose enters these cells after a meal, its metabolism increases the ATP-to-ADP ratio, closing ATP-sensitive potassium channels, depolarizing the cell membrane, and opening voltage-gated calcium channels; calcium influx then triggers insulin secretion. Insulin promotes glucose uptake and storage in tissues, helping maintain metabolic balance. Studying beta-cell development, function, dysfunction, and survival supports research into diabetes, including mechanisms of insulin deficiency and resistance, as well as the development of cell-based therapies, drug treatments, and strategies for pancreatic regeneration.

Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

The present protocol describes an enhanced method to increase the co-expression of PDX1 and NKX6.1 transcription factors in pancreatic progenitors derived from human pluripotent stem cells (hPSCs) in planar monolayers.
