Plastic Embedding Sectioning

Plastic embedding sectioning is a tissue-preparation technique that encases biological specimens in a hardened plastic resin so they can be cut into thin, stable sections for microscopic examination. After fixation and dehydration, the specimen is infiltrated with resin, which polymerizes around the tissue; a microtome then produces sections that can be mounted and stained for imaging. This approach preserves cellular architecture, supports analysis of hard or delicate samples, and enables high-resolution studies in histology, developmental biology, pathology, and microscopy. By maintaining structural detail during sectioning, plastic embedding helps researchers relate tissue organization to biological function and disease-related changes.

Plastic sections maintain true tissue morphology in thin sections of tissue that can be immunostained with fluorescent secondary antibodies, making this method more useful than paraffin-embedded or frozen sections for many types of tissue. The method for staining, plastic embedding, and sectioning is demonstrated in this video.

Identification of mechanisms underlying muscle damage is crucial. Here we present the histological technique for preparing paraffin-embedded and frozen sections of Drosophila thoracic muscles. This allows analysis of muscle morphology and localization of protein and other muscle cell components.

Pancreatic Islet Embedding for Paraffin Sections

Ex vivo pancreatic islet studies are important for diabetes research. Existing techniques to study cultured islets in their native 3-dimensional architecture are time consuming, inefficient, and infrequently used. This work describes a new, simple, and efficient method for generating high-quality paraffin sections of whole cultured islets.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
