Ploidy Determination

Ploidy determination is the assessment of how many complete sets of chromosomes a cell or organism contains, distinguishing haploid, diploid, and polyploid states. In biology, it commonly relies on measuring DNA content or examining chromosome number through methods such as flow cytometry, fluorescence microscopy, or chromosome counting; DNA-binding dyes produce signals that correlate with genome copy number. This analysis helps characterize cell division, development, reproduction, and genome stability, while identifying aneuploidy or polyploidy associated with genetic disorders, cancer, plant breeding, and evolutionary change. Accurate ploidy measurements also support research involving cell cultures, hybrid organisms, and genome manipulation.

Manipulation of Ploidy in Caenorhabditis elegans

This method allows for the generation of tetraploid and triploid Caenorhabditis nematodes from any diploid strain. Polyploid strains generated by this method have been used to study chromosome interactions in meiotic prophase, and this method is useful for examining important basic questions in cell, developmental, evolutionary, and cancer biology.

Oocytes are prone to aneuploidy due to errors in chromosome segregation during meiotic maturation. Aneuploid eggs can cause infertility, miscarriages or developmental disorders like Down syndrome. Here, we describe methods to introduce materials of choice into oocytes and methods to study meiotic maturation and assess ploidy.

A modified protocol for ploidy manipulation uses a heat shock to induce a one-cycle stall in cytokinesis in the early embryo. This protocol is demonstrated in the zebrafish but may be applicable to other species.

A High-Throughput In Situ Method for Estimation of Hepatocyte Nuclear Ploidy in Mice

We present a robust, cost-effective, and flexible method for measuring changes in hepatocyte number and nuclear ploidy within fixed/cryopreserved tissue samples that does not require flow cytometry. Our approach provides a powerful sample-wide signature of liver cytology ideal for tracking the progression of liver injury and disease.
