Proto Oncogenes

Proto-oncogenes are normal genes that regulate cell growth, division, differentiation, and survival, helping maintain healthy tissue development and function. They typically encode growth factors, cell-surface receptors, intracellular signaling proteins, or transcription factors that transmit signals controlling gene expression and the cell cycle. Mutations, gene amplification, or chromosomal rearrangements can abnormally activate a proto-oncogene, converting it into an oncogene that drives uncontrolled proliferation and contributes to cancer. Studying these genes helps researchers understand tumor initiation and progression, identify molecular biomarkers, and develop targeted therapies that interrupt specific growth-signaling pathways.

Cancer-Critical Genes I: Proto-oncogenes

2023

Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype. When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors. Such genes that act...

Mapping the Structure-Function Relationships of Disordered Oncogenic Transcription Factors Using Transcriptomic Analysis

Intrinsically disordered domains are important for oncogenic fusion transcription factor function. To therapeutically target these proteins, a more detailed understanding of the regulatory mechanisms employed by these domains is required. Here, we use transcriptomics to map important structural features of the intrinsically disordered EWS domain in Ewing sarcoma.
