Rfp-ara7

RFP-ARA7 is a red fluorescent protein fusion used to visualize ARA7, an Arabidopsis Rab5-like GTPase associated with endosomal trafficking. The fluorescent tag follows ARA7 as it associates with endosomal membranes, allowing researchers to identify and monitor pre-vacuolar compartments and multivesicular bodies in living plant cells. In biology, RFP-ARA7 supports studies of membrane transport, protein sorting, and vacuole-directed trafficking by providing a visible marker for endosomal compartments. Imaging this reporter can help reveal how vesicles form, move, and mature, offering insight into plant cell organization and the cellular pathways that regulate cargo delivery.

We describe a novel red fluorescent protein (RFP) reporter that is expressed specifically in the Drosophila eye. We detail a methodology for dissection of the eye imaginal disc and how this reporter can be used to aid in the dissection and identification of specific cell types in the developing eye.

In vitro spheres assays are commonly used to identify cancer stem cells. Here we compare single with multi cell-based spheres assays. The more laborious single cell-based assays or methylcellulose supplementation give more accurate results while multi cell-based assays performed in liquid medium can be highly influenced by cell density.
