Rna Dimerization

RNA dimerization is the association of two RNA molecules into a higher-order complex, a process that can alter RNA structure, stability, and biological activity. It occurs through complementary base pairing or specific noncanonical interactions, often involving exposed loops, stems, and tertiary contacts that bring separate RNA strands together under suitable ionic and structural conditions. In biology, RNA dimerization contributes to viral genome packaging, RNA regulation, and the assembly of ribonucleoprotein complexes. Studying these interactions helps explain how RNA molecules organize genetic information and supports the design of RNA-based therapeutics, sensors, and molecular tools.

Isolating Free Carbenes, their Mixed Dimers and Organic Radicals

We present protocols for the isolation of stable heterocyclic carbenes. The synthesis of a cyclic (alkyl)(amino) carbene (CAAC) and an N-heterocyclic carbene (NHC) is demonstrated using filter cannulas and Schlenk technique. We furthermore present the synthesis of the related oxygen-sensitive, electron-rich mixed “Wanzlick dimer” and the reduced stable organic radical.

Chemical Dimerization-Induced Protein Condensates on Telomeres

This protocol illustrates a chemically induced protein dimerization system to create condensates on chromatin. The formation of promyelocytic leukemia (PML) nuclear body on telomeres with chemical dimerizers is demonstrated. Droplet growth, dissolution, localization and composition are monitored with live cell imaging, immunofluorescence (IF) and fluorescence in situ hybridization (FISH).

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.
