Synovial Fluid Exosomes

Synovial fluid exosomes are nanoscale extracellular vesicles released by cells within joints and transported through the fluid that lubricates and supports articular tissues. Their lipid bilayer protects proteins, lipids, and nucleic acids, allowing these cargoes to enter recipient cells and influence processes such as inflammation, matrix turnover, and cell communication. In biology and musculoskeletal research, analyzing exosome composition can help clarify communication between chondrocytes, synovial cells, and immune cells during joint maintenance or disease. Their molecular cargo also supports investigation of biomarkers and potential vehicles for targeted therapeutic delivery in conditions such as osteoarthritis.

A 3D System for Culturing Human Articular Chondrocytes in Synovial Fluid

A 3D system of culturing human articular chondrocytes in high levels of synovial fluid is described. Synovial fluid reflects the most natural microenvironment for articular cartilage, and can be easily obtained and stored. This system thus can be used for studying cartilage regeneration and for screening therapeutics for treating arthritis.

A protocol establishing mesenchymal stem cells (MSCs) isolated from the synovial fluid and bone marrow of minipigs and non-invasively collected using syringe aspiration is presented. Cellular phenotyping was performed using flow cytometry after cell isolation and in vitro cultivation of bone marrow and synovial fluids derived from MSCs.
