Three-dimensional Assay

Three-dimensional assays are biological research methods that measure cellular behavior or molecular activity in models organized within a three-dimensional environment rather than on a flat culture surface. They typically embed cells in extracellular-matrix materials or assemble them as spheroids, allowing cell-cell and cell-matrix interactions, nutrient and oxygen gradients, and tissue-like architecture to influence experimental readouts. In biology, these assays can assess proliferation, differentiation, invasion, drug response, and organoid development under conditions that more closely represent living tissues. By capturing spatial effects that two-dimensional assays may miss, they support disease modeling, therapeutic evaluation, and the development of more physiologically relevant experimental systems.

A method is described for the preparation of a 3-dimensional matrix consisting of collagen type I and primary human fibroblasts. This organotypic gel serves as a useful substrate to assess invasive cell migration because it mimics basic features of tissue stroma and is amenable to many forms of microscopy.
