Tunica Albuginea

The tunica albuginea is a dense, collagen-rich connective tissue capsule that surrounds certain organs, most notably the testis and ovary. In the testis, it forms a protective outer layer and sends fibrous septa inward, dividing the organ into lobules that contain seminiferous tubules; in the ovary, it lies beneath the surface epithelium and contributes to the cortex’s structural organization. Its thickness, collagen composition, and relationship to surrounding tissue can change with development, injury, or disease. Histological study of the tunica albuginea helps interpret gonadal architecture and investigate conditions affecting spermatogenesis, ovarian tissue, or organ integrity.

This article introduces a surgical method using a pedicled tunica vaginalis to treat long-segment urethral stricture resulting from lichen sclerosus. The tunica vaginalis is an excellent substitute for the urethra, leading to quick postoperative recovery for patients.

Primary Sertoli cells are required for studying testis-immune privilege, signal transduction during inflammation or infection, and utilization of their immunoprotective properties. Here we describe an enzyme-based protocol for the isolation of highly purified primary Sertoli cells and peritubular cells from rat testes.

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.
