Undecalcified Bone Histology

Undecalcified bone histology is the microscopic examination of bone that retains its native mineral content, preserving the tissue’s structure and mineralized matrix for accurate analysis. Because decalcification is omitted, specimens are fixed, dehydrated, embedded in a hard resin, and sectioned or ground into thin slices before staining or microscopy; fluorescent labels can reveal the timing of new mineral deposition. This method supports the study of bone formation, resorption, remodeling, and mineralization in biology and biomedical research. It is particularly valuable for evaluating skeletal development, metabolic bone disease, osteoporosis, and the integration of implants with surrounding bone.

Undecalcified bone histology provides important information for a variety of clinical and research applications. It is technically challenging, particularly with large size specimens. This video illustrates the process of producing good quality sections and demonstrates the technical difficulties and methods with which to overcome them.

Automated Quantification of Hematopoietic Cell – Stromal Cell Interactions in Histological Images of Undecalcified Bone

A strategy to quantitatively analyze histological data in the bone marrow is presented. Confocal microscopy of fluorescently labeled cells in tissue sections results in 2-dimensional images, which are automatically analyzed. Co-localization analyses of different cell types are compared to data from simulated images, giving quantitative information about cellular interactions.

Animal models are frequently utilized to study cancer metastasis to bone. In this protocol we will describe two common methods of tumor inoculation for bone metastasis studies and briefly describe some of the analyses utilized to monitor and quantify these models.
