Wick Preparation Technique

The Wick Preparation Technique is a method for preparing a porous material that transports liquids through capillary action, making it useful for controlled fluid handling in biological experiments. A prepared wick draws solution into its fibers or pores when surface tension and adhesion overcome gravity, allowing liquid movement without an external pump; its performance depends on material properties, dimensions, and contact with the solution. In biology, wick-based setups can support sample hydration, reagent delivery, filtration, and simple microfluidic workflows. Careful preparation promotes consistent flow and helps researchers create accessible experimental systems for studying transport, reactions, and cellular responses.

Source: Lin, P., et al. Synthesizing Sodium Tungstate and Sodium Molybdate Microcapsules via Bacterial Mineral Excretion. J. Vis. Exp. (2018).This video demonstrates the preparation of a monoclonal bacterial culture for mineral production through serial dilution and colony isolation on an agar plate. By shaking the plate with glass beads, individual cells are spatially separated to form genetically uniform colonies, which are then transferred to liquid media for growth. The culture with the...

Sample Preparation for Analysis: Advanced Techniques

2024

Accurate analysis of complex samples often requires advanced preparation techniques to achieve reliable and reproducible results. Samples containing inorganic or organic materials can be challenging to dissolve or decompose effectively. Standard sample preparation methods include acid digestion, fusion, dry ashing, and wet digestion. Acid digestion with strong acids is commonly used to dissolve inorganic materials that are insoluble (do not dissolve) in water. This method can be useful for...

Here we describe a protocol to prepare and maintain primary pituitary cell cultures from medaka (Oryzias latipes). The optimized conditions in this protocol take important parameters such as temperature, osmolality, and pH into consideration by mimicking the physiological conditions of the fish, thereby enabling physiologically more meaningful results.

The preparing and processing of dried blood spots (DBS) for their final analysis are still poorly standardized for most diagnostic applications. To overcome this shortcoming, a comprehensive step-by-step protocol is suggested and subsequently evaluated with regard to its effectiveness for detecting markers of viral infections.
