Granule Cell Precursors

Granule cell precursors are proliferative neural progenitor cells that generate cerebellar granule neurons during development, making them important for studying brain formation and tumors derived from neural lineages. They expand in the cerebellar external germinal layer when Sonic hedgehog signaling activates GLI-dependent transcription, then migrate inward and differentiate as signaling declines. In cancer research, abnormal pathway activation can maintain precursor-like proliferation and contribute to medulloblastoma, particularly the SHH molecular subgroup. Their culture and genetic manipulation support studies of tumor initiation, developmental signaling, therapeutic vulnerabilities, and treatment response, linking normal neurogenesis to cancer biology.

Ex Vivo Culture of Chick Cerebellar Slices and Spatially Targeted Electroporation of Granule Cell Precursors

The cerebellar external granule layer is the site of the largest transit amplification in the developing brain. Here, we present a protocol to target genetic modification to this layer at the peak of proliferation using ex vivo electroporation and culture of cerebellar slices from embryonic Day 14 chick embryos.

Investigating Mast Cell Secretory Granules; from Biosynthesis to Exocytosis

The goal of the present protocol was to develop a method that will allow functional genomic analyses of mast cell secretion. The protocol is based on quantitative assessment of the release of a fluorescent reporter gene cotrasfected with the gene of interest and real time analyses of the secretory granule's morphology.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.

During postnatal cerebellum development, immature granule cells originating from the germinal zone exhibit distinct modalities of migration to reach their final destination and to establish neuronal networks. This protocol describes the preparation of cerebellar slices and the confocal macroscopic approach used to investigate the factors that regulate neuronal migration.
