Mrna Electroporation

mRNA electroporation is a technique that uses brief electrical pulses to introduce messenger RNA into cells, enabling temporary protein production without altering the genome. The electric field creates transient pores in the cell membrane, allowing mRNA to enter the cytoplasm, where ribosomes translate it into a selected protein before the RNA is naturally degraded. In cancer research, this approach can program immune cells to express tumor-associated antigens, receptors, or immune-modulating proteins for studying antitumor responses and developing cell-based therapies. Its transient, nonviral delivery profile supports rapid testing of therapeutic designs while limiting permanent genetic changes.

We report messenger RNA (mRNA) electroporation as a method that permits fast and efficient expression of multiple proteins in the quail embryo model system. This method can be used to fluorescently label cells and record their in vivo movements by time-lapse microscopy shortly after electroporation.
