Preclinical Cancer Testing

Preclinical cancer testing is the evaluation of cancer therapies before they enter human clinical trials, helping researchers assess potential benefits, risks, and biological activity. It combines laboratory models, such as cultured tumor cells and tumor-bearing animals, with controlled experiments that measure drug response, tumor growth, molecular effects, pharmacokinetics, and toxicity. In cancer research, these studies help clarify how a treatment acts on cancer cells and surrounding tissues, identify effective doses and treatment combinations, and determine whether results justify clinical development. Although preclinical findings do not guarantee patient benefit, rigorous testing can improve trial design, reveal limitations early, and support more informed therapeutic decisions.

This method utilizes zebrafish embryos to efficiently test the vascular invasive ability of cancer cells. Fluorescent cancer cells are injected into the precardiac sinus or yolk sac of developing embryos. Cancer cell vascular invasion and extravasation is assessed via fluorescence microscopy of the tail region 24 to 96 hr later.

Patient-derived xenograft (PDX) models and transplantable genetically engineered mouse models faithfully recapitulate human disease and are preferred models for basic and translational breast cancer research. Here, a method is described to orthotopically transplant breast tumor fragments into the mammary fat pad to study tumor biology and evaluate drug response.
