Slc Superfamily

The SLC superfamily is a large group of membrane transport proteins that move ions, metabolites, nutrients, and drugs across cell membranes, making it essential for cellular homeostasis and human physiology. SLC proteins function through facilitated diffusion, secondary active transport, or exchange, using concentration and electrochemical gradients to determine substrate movement. In clinical research, altered SLC expression or activity can contribute to inherited disorders, cancer, neurological disease, and drug resistance. Characterizing these transporters supports disease-mechanism studies, biomarker development, and pharmacology by helping predict drug uptake, distribution, interactions, and treatment response.

Creating and Applying a Reference to Facilitate the Discussion and Classification of Proteins in a Diverse Group

2017

The goal of this protocol is to develop a reference for divergent proteins in a group that lacks coherent criteria for nomenclature and classification. This reference will facilitate analyses and discussion of the group as a whole and can be used in addition to established names.

2014

The zebrafish embryo is an excellent model for developmental biology research. During embryogenesis, zebrafish develop with a yolk mass, which presents three-dimensional challenges for sample observation and analysis. This protocol describes how to create two-dimensional flat mount preparations of whole mount in situ (WISH) stained zebrafish embryo specimens.

Protein Families

2020

Protein families are groups of homologous proteins; that is, they have similarities in amino acid sequences and three-dimensional structures. Protein families usually occur because of gene duplication, where an additional copy of a gene is inserted into the genome of an organism. Mutations that change the amino acids but still allow the protein to be properly synthesized, will lead to new protein family members. If these new proteins contain similar amino acids in key locations, protein...

Transduction of Human Cells with Polymer-complexed Ecotropic Lentivirus for Enhanced Biosafety

2011

Lentiviruses are a valuable research tool for exploring gene function; however, researchers may wish to avoid production of pantropic lentivirus encoding known or suspected oncogenes. As an alternative, we present a safer protocol for use of ecotropic lentivirus on human cells modified to express the ecotropic receptor mSlc7a1.
