Cryopreservation Intermediate Banks

Cryopreservation intermediate banks are organized collections of frozen biological materials maintained between initial sample preparation and downstream experimental use, supporting consistent access to developmental biology specimens. Samples are combined with cryoprotective agents, cooled under controlled conditions to limit ice-crystal formation, and stored at ultra-low temperatures, typically in liquid nitrogen, to preserve cellular structure and viability. These banks help standardize studies by providing traceable, reproducible stocks of cells, tissues, embryos, or other developmental materials. They also reduce sample loss, support longitudinal experiments, and facilitate collaboration by enabling well-characterized biological resources to be distributed and recovered when needed.

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.
