Mesenchymal Cell Preservation

Mesenchymal cell preservation is the controlled storage of mesenchymal stromal cells to maintain their viability, identity, and functional properties for later use in developmental biology and regenerative research. Cryopreservation typically uses a protective medium containing cryoprotectants and controlled cooling to limit ice-crystal formation, followed by storage at very low temperatures and carefully regulated thawing. Proper preservation supports consistent cell availability for studying differentiation, tissue development, cell signaling, and interactions with developing tissues. It also helps researchers compare experiments across time, reduce culture-related variation, and prepare standardized cell populations for applications in tissue engineering and cell-based therapies.

Isolating and Analyzing Cells of the Pancreas Mesenchyme by Flow Cytometry

Here, we describe a method for the isolation of cells in the pancreas microenvironment from embryonic, neonatal and adult mouse tissue, focusing on the isolation of mesenchymal cells. This method allows profiling of cell gene expression and protein secretion in order to elucidate the extrinsic signals that regulate pancreas development, function, and tumorigenesis.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2). This method is suitable for better understanding the biological impact of phenotypic plasticity on cancer aggressiveness and treatments.
