Blunt-end Micropipette

A blunt-end micropipette is a glass microtool with a flattened, nonpointed tip used to aspirate, hold, or transfer cells and other biological material with controlled contact. Connected to a micromanipulator and pressure system, it regulates fluid movement through the pipette while its blunt geometry supports gentle handling and reduces the risk of puncturing or cutting delicate samples. In genetics, blunt-end micropipettes are valuable for positioning oocytes and embryos, transferring cells, and supporting microinjection or biopsy workflows alongside sharper injection pipettes. Their precise handling improves sample integrity and helps researchers perform reproducible genetic manipulation and analysis.

Micropipette Aspiration of Substrate-attached Cells to Estimate Cell Stiffness

Here we describe a quick and simple method to measure cell stiffness. The general principle of this approach is to measure membrane deformation in response to well-defined negative pressure applied through a micropipette to the cell surface. This method provides a powerful tool to study biomechanical properties of substrate-attached cells.

A Scalable Model to Study the Effects of Blunt-Force Injury in Adult Zebrafish

We modified the Marmarou weight drop model for adult zebrafish to examine a breadth of pathologies following blunt-force traumatic brain injury (TBI) and the mechanisms underlying subsequent neuronal regeneration. This blunt-force TBI model is scalable, induces a mild, moderate, or severe TBI, and recapitulates injury heterogeneity observed in human TBI.

We introduce the Fluorescence Micropipette Aspiration (fMPA) technique, a novel method combining controlled mechanical stimulation with comprehensive analysis of intracellular signaling in single cells. Bacterial mechanosensitive channels can be used as mechanoelectrical transducers in biomolecular devices. Droplet interface bilayers (DIBs), cell-inspired building blocks to such devices, represent new platforms to incorporate and stimulate mechanosensitive channels. Here, we demonstrate a new micropipette-based method of forming DIBs, allowing the study of mechanosensitive channels under mechanical stimulation.
