Gentamicin Protection Assay

The gentamicin protection assay is a microbiological technique used to quantify bacteria that enter and survive inside cultured host cells, making it valuable for studying cellular infection. In the assay, extracellular bacteria are exposed to gentamicin, an antibiotic that generally does not penetrate intact eukaryotic cells effectively; after antibiotic treatment, host cells are washed, lysed, and plated to measure surviving intracellular bacteria as colony-forming units. Researchers use this method to assess bacterial invasion, intracellular survival, and the effects of host-cell responses, antimicrobial treatments, or bacterial mutations in immunology and infection studies.

Antibiotic resistance amongst adherent-invasive Escherichia coli strains poses challenges for assessing bacterial invasion using the traditional gentamicin protection assay. An improved method to evaluate bacterial invasion quantitatively by enumerating colony-forming units and qualitatively through visualization of infected cells by immunofluorescence confocal microscopy is described.

Kidney Regeneration in Adult Zebrafish by Gentamicin Induced Injury

Here we present a reliable method to study adult kidney regeneration by inducing acute kidney injury by gentamicin injection. We show that injury is dependent on gentamicin dosage and environmental temperature using in situ hybridization to label lhx1a+ developing new nephrons.
