Immune Cell Identification

Immune cell identification is the classification of leukocytes and their functional states using distinctive cellular features, a foundational step in immunology and infection research. Researchers distinguish cells through morphology, lineage-specific surface markers, intracellular proteins, and patterns of gene or cytokine expression, often using flow cytometry, immunostaining, or microscopy. These methods can identify populations such as T cells, B cells, macrophages, and neutrophils while revealing activation, differentiation, or infection-associated changes. Accurate identification supports diagnosis, pathogen-response studies, vaccine development, and evaluation of immune-targeting therapies by linking specific cell populations to disease mechanisms and treatment outcomes.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
