Immune Cell Purity

Immune cell purity is the proportion of a cell preparation composed of the intended immune cell population rather than unwanted cells, debris, or microbial contaminants. Researchers assess and improve purity by separating cells with properties such as density, surface-marker expression, or antibody binding, then validating composition through methods including flow cytometry or microscopic analysis. High-purity preparations are essential for interpreting experiments accurately because contaminating cells can alter cytokine measurements, gene-expression profiles, and functional responses. In immunology and infection research, purity assessment supports reliable studies of host-pathogen interactions, immune-cell signaling, diagnostics, therapeutic development, and cell-based applications.

Determination of Immune Cell Identity and Purity Using Epigenetic-Based Quantitative PCR

Here we describe a robust method of determining immune cell identity and purity through epigenetic signatures detected using quantitative PCR (qPCR). DNA demethylation at a specific locus serves as a unique identifier for a particular cell type and allows for identification of CD8+, regulatory, or Th17 T cells.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
