Ovalbumin-specific Receptors

Ovalbumin-specific receptors are immune recognition molecules that bind ovalbumin or peptides derived from this egg-white protein, enabling study of antigen-specific responses. In experimental immunology, ovalbumin peptides are processed by antigen-presenting cells and displayed on major histocompatibility complex molecules; antigen receptors, particularly T-cell receptors, recognize the resulting peptide–MHC complexes and activate signaling pathways when binding is sufficient. These receptor systems provide defined models for examining T-cell activation, clonal expansion, immune tolerance, and inflammatory responses. In infection research, ovalbumin-specific responses can serve as controlled tools for evaluating antigen presentation, vaccine adjuvants, immunotherapies, and mechanisms that regulate host defense.

This video demonstrates a technique for the ex vivo generation of tumor antigen-specific chimeric antigen receptor (CAR) T cells. Retroviral vectors carrying transgenic RNA are mixed with spleen cells in a medium supplemented with interleukin-2 to promote T-cell survival and proliferation. We demonstrate here that epigenetic reprogramming via Somatic Cell Nuclear Transfer (SCNT) can be used as a tool to generate mouse models with pre-defined T cell receptor (TCR) specificities. These transnuclear mice express the corresponding TCR from their endogenous locus under the control of the endogenous promoter.

Herein we describe a novel method to generate antigen-specific T cell receptors (TCRs) by pairing the TCRα or TCRβ of an existing TCR, possessing the antigen-specificity of interest, with complementary hemichain of the peripheral T cell receptor repertoire. The de novo generated TCRs retain antigen-specificity with varying affinity.
